Hydrochlorothiazide works by inhibiting the sodium-chloride symporter in the distal convoluted tubule of the nephron, which increases the excretion of sodium, chloride, and water, thereby reducing blood volume and blood pressure. Hydrochlorothiazide acts on the proximal region of the distal convoluted tubule, inhibiting reabsorption by the sodium-chloride symporter.
